Conclusion

The developed black membrane slide provides an outstanding detectable concentration range in fluorescence analysis of proteins. The low background signal of the black membrane slide allows to collect signal intensity not only from the surface but also from the depth of the microporous structure. The 3D structure was designed to offer a high binding affinity for proteins and to better stabilize their active conformation even in presence of denaturing temperature conditions. As a consequence the dynamic range of the developed slide is increased and the experimental protocol is more robust than that one of conventional 2D and white 3D substrates.
